Is there any evidence for any medication in the treatment of Peyronie's disease, including Neprinol and Vitamin E?

ATTRACT, the Welsh equivalent of the NLH Q&A Service, answered a question on Peyronie's disease and Vitamin E [1] – see reference section for more information.  Since that answer we have been unable to find any more trials so that answer appears ‘current’.

With respect to neprinol we have found no clinical trials. 

In 2003 NICE issued guidance on extracorporeal shockwave therapy [2] but that is hardly medication.

With regard to other medications (using the term broadly) we searched Medline and the Cochrane Library for RCTs published since 2000.  We found a number and as such we’ll simply report the title and conclusions.  The link to the abstract is given in the reference section.  NOTE: we have not assessed these trials for bias.

Title: Effect of intralesional interferon-alpha 2b combined with oral vitamin E for treatment of early stage Peyronie's disease: a randomized and prospective study. [3]
Conclusions: Our findings indicate that 5 million units of intralesional interferon-alpha 2b injection therapy either alone or in combination with vitamin E does not appear to be clinically effective in the management of early stage Peyronie's disease compared with only oral vitamin E.

Title: Liposomal recombinant human superoxide dismutase for the treatment of Peyronie's disease: a randomized placebo-controlled double-blind prospective clinical study [4]
Conclusion: LrhSOD is an easily administrable, safe and effective local therapeutic for the painful phase of Peyronie's Disease.

Title: Potassium paraaminobenzoate (POTABA) in the treatment of Peyronie's disease: a prospective, placebo-controlled, randomized study. [5]
Conclusion: The results of this study indicate a significant plaque-related effect of potassium paraaminobenzoate. There was no relevant difference with regard to improvement of pre-existing penile deviation. However, under potassium paraaminobenzoate a significant protective effect on deterioration of penile curvature could be demonstrated. Potassium paraaminobenzoate appears to be useful to stabilize the disorder and prevent progression of penile curvature.

Title: A prospective, randomized study using transdermal electromotive administration of verapamil and dexamethasone for Peyronie's disease. [6]
Conclusion: Intraplaque electromotive verapamil and dexamethasone induce substantial objective improvement in Peyronie's disease compared to electromotive lidocaine administration.

Title. Therapeutic effects of colchicine in the management of Peyronie's disease: a randomized double-blind, placebo-controlled study. [7]
Conclusion: Colchicine is no better than placebo in improvement of pain, curvature angle, or plaque size in patients with Peyronie's disease.

Title: Oral propionyl-l-carnitine and intraplaque verapamil in the therapy of advanced and resistant Peyronie's disease. [8]
Conclusion: The combination of propionyl-l-carnitine and verapamil can be considered the therapy of choice for advanced and resistant Peyronie's disease.

Title: Acetyl-L-carnitine vs tamoxifen in the oral therapy of Peyronie's disease: a preliminary report [9]
Conclusion: These results suggest that acetyl-L-carnitine is significantly more effective and safe than tamoxifen in the therapy of acute and early chronic Peyronie's disease.

Title: The impact of intralesional interferon alpha-2b injection therapy on penile hemodynamics in men with Peyronie's disease. [10]
Conclusion: This study reveals that intralesional IFN alpha-2b injections have a significant benefit on penile hemodynamic parameters. Moreover, intralesional IFN alpha-2b is an effective, minimally invasive treatment for PD, and PDDU is a useful adjunct to monitor objectively changes in penile vascular parameters. interferon alpha-2b injection therapy on penile hemodynamics in men with Peyronie's disease.
